Pharmacodynamics:

A selective cyclooxygenase inhibitor, an enzyme that catalyzes the reaction of the conversion of arachidonic acid to prostaglandin H2, which is the precursor of the remaining prostaglandins, thromboxane A2 and prostacyclin. Has anti-inflammatory and analgesic, as well as antipyretic effect.

Increases the resistance of cells, stimulating the healing of wounds. Influences on the center of thermoregulation, having an antipyretic effect. Stimulates the synthesis and production of interferon.

Pharmacokinetics:

After oral administration, up to 98% is absorbed in the gastrointestinal tract. The maximum concentration in the blood plasma is reached after 2-4 hours. The connection with plasma proteins is up to 80%.

Therapeutic effect develops after the reception. Metabolism in the liver.

The elimination half-life is 3 hours. Elimination by the kidneys and with feces, 67% - in unmodified form.

Indications:

It is used for rheumatism, arthralgia, myalgia, headache and toothache, neuralgia and fever.

Contraindications:

Peptic ulcer of stomach and duodenum, ulcerative colitis, gastritis.

Individual intolerance.

Pregnancy and lactation:

Dosing and Administration:

Application in children

5-10 years: 0.25 g 3-4 times / day.

10-18 years: 0.3 g / day.

Adults

Inside, after eating 0.25-0.5 g 3-4 times / day.

The highest daily dose: 3 g.

The highest single dose: 0.5 g.

Side effects:

Nausea, heartburn, diarrhea.

Allergic reactions.

Overdose:

Symtomas: epigastric pain, nausea, vomiting, drowsiness, rarely - gastrointestinal bleeding, muscle twitching, respiratory depression, coma.

Treatment: symptomatic therapy.

Interaction:

Increases the effectiveness of oral hemocoagulants.

With simultaneous use with barbiturates, narcotic analgesics, caffeine, phenothiazine derivatives, diphenhydramine, phenacetin, analgesic effects are enhanced.

The anti-inflammatory effect of the drug is enhanced by salicylates and metamizol sodium.
