    Pharmacodynamics:

    Latanoprost

    The analogue of prostaglandin F2α, a selective agonist of FP-receptors. Increases the outflow of moisture along the uveoscleral pathway, reducing intraocular pressure. The effect of the drug begins 3-4 hours after the administration, the maximum effect develops after 8-12 hours. The duration of action is 24 hours.

    Timolol

    It increases the outflow of the uveoscleral outflow, reduces the secretion of moisture by the cells of the ciliated epithelium. Non-selective β1 and β2 adrenoblocker, which does not have significant sympathomimetic activity. Does not have a depressive effect on the myocardium, does not have membrane-stabilizing activity.

    Pharmacokinetics:

    Latanoprost

    It freely penetrates through the cornea, hydrolyses to a biologically active form. The maximum concentration in aqueous humor occurs 2 hours after topical application.

    The half-life is 17 min.

    Timolol

    The maximum concentration in aqueous humor is reached after 1 hour.Part of the dose is absorbed into the blood plasma and has a systemic effect.

    The half-life is 6 hours. It is metabolized in the liver. Elimination by the kidneys.

    Indications:

    It is used to reduce intraocular pressure in patients with elevated ophthalmotonus and open-angle glaucoma.

    Contraindications:
    • Bronchial asthma (including history).
    • Chronic heart failure of II and III degree.
    • Atrioventricular blockade of II-III degree.
    • Cardiogenic shock.
    • Individual intolerance.
    Carefully:

    Afakia, pseudo-aphakia. Congenital, inflammatory, neovascular forms of glaucoma. Diabetes mellitus, thyrotoxicosis, myasthenia gravis.

    Pregnancy and lactation:

    Recommendations for FDA - Category C. Contraindicated in pregnancy. When treating the drug, it is recommended to stop breastfeeding.

    Dosing and Administration:

    In the morning 1 drop every 1 day in the conjunctival sac.

    The highest daily dose: 2 drops.

    The highest single dose: 2 drops.

    Side effects:

    Latanoprost

    Eye irritation: itching, sensation of foreign body, hyperemia of the conjunctiva; increased pigmentation of the iris, macular edema, blurred vision.

    Timolol

    Keratitis, diffraction disorders, diplopia, ptosis, tinnitus.

    Allergic reactions.

    Overdose:

    Latanoprost

    Hyperemia of the eyes and eyelids.

    Timolol

    Bradycardia, arterial hypotension, bronchospasm, cardiac arrest.

    Treatment is symptomatic.

    Interaction:

    Means of reducing catecholamine activity, slow calcium channel blockers, antiarrhythmics, cardiac glycosides digitalis group, β-blockers, narcotic analgesics, anticholinergic and monoamine oxidase inhibitors, when used together amplify hypotensive effect and lead to the development of bradycardia.

    Monitoring of the patient's condition.

    Before using the drug, the patient should be warned about irreversible changes in eye color, darkening of the eyelid skin, changes in thickness, length and number of eyelashes.

    Persons using contact eye correction should wear the lenses no earlier than 15 minutes after instillation.
