The mechanism of action is due to the direct influence of gopantenic acid on GABA_B-receptor-channel complex. The drug has nootropic and anticonvulsant action. Increases brain resistance to hypoxia and the effects of toxic substances, stimulates anabolic processes in neurons,combines a moderate sedative effect with a mild stimulating effect, has an anticonvulsant effect, reduces motor excitability. Increases mental and physical performance. Helps normalize GABA content in chronic alcohol intoxication and subsequent ethanol elimination. Prolongs the effect of novocaine and sulfonamides by inhibiting the reactions of their acetylation. It causes inhibition of the pathologically increased vesicle reflex and detrusor tone.

Pharmacokinetics:

Quickly absorbed from the gastrointestinal tract, penetrates the blood-brain barrier, the greatest concentrations are created in the liver, kidneys, in the wall of the stomach and skin. Time to reach C_mOh- 1 hour. The drug is not metabolized and is excreted unchanged for 48 hours: 67.5% of the dose taken with urine, 28.5% with feces.

Indications:

- Psycho-emotional overload, reduction of mental and physical performance (increased concentration and memorization).

As part of complex therapy:

- Cognitive disorders in organic brain lesions (including number of consequences of neuroinfections and craniocerebral injuries);

- schizophrenia with cerebral organic insufficiency;

- cerebrovascular insufficiency caused by atherosclerotic changes in cerebral vessels;

- extrapyramidal disorders (myoclonus-epilepsy, Huntington's chorea, hepatolenticular degeneration, Parkinson's disease, etc.);

- epilepsy with a delayed mental processes, together with anticonvulsant drugs;

- neurogenic disorders of urination: enuresis, pollakiuria, imperative urges, imperative urinary incontinence;

- in children with perinatal encephalopathy, mental retardation of varying severity, with developmental delay (mental, speech, motor or combination), with various forms of infantile cerebral palsy, hyperkinetic disorders (attention deficit hyperactivity disorder, tics), stuttering clonic form), neurosis-like states.

The drug is used in children older than 3 years. At an earlier age, it is recommended that the drug be administered as a syrup.

Contraindications:

- Hypersensitivity;

- Acute severe kidney disease;

- Pregnancy (I trimester);

- Children under 3 years old.

Dosing and Administration:

Calcium gopantenate is used alone or as part of complex therapy.

The drug is taken orally 15-30 minutes after eating.

Single dose for adults - 0.25-1 g, for children - 0,25-0,5 g, daily dose for adults - 1,5-3 g, for children - 0,75-3 g. The course of treatment lasts from 1 to 4 months, in some cases up to 6 months. After 3-6 months, a repeat course of treatment is possible.

Application of the drug in monotherapy:

To restore working capacity under increased loads and asthenic conditions the drug is prescribed to 0.25 g 3 times a day.

As part of complex therapy:

- epilepsy the drug is prescribed to children 0.25-0.5 g 3-4 times a day, adults 0.5-1 g 3-4 times a day, daily, for a long time (up to 6 months);

- hyperkinesis the drug is prescribed to children 0.25-0.5 g 3-6 times daily for 1-4 months, adults - 1.5-3 g daily for 1-5 months;

- in children with mental deficiency and oligophrenia the drug is prescribed for 0.5 g 4-6 times a day, daily for 3 months, with delay speech development - 0.5 g 3-4 times a day for 2-3 months;

- consequences of neuroinfections and craniocerebral injuries the drug is prescribed 0.25 g 3-4 times a day;

- ticks the drug is prescribed to children 0.25-0.5 g 3-6 times a day, 1-4 months, adults 1.5-3 g per day, for 1-5 months;

- disorders of urination the drug is prescribed 2-3 times a day for adults to 0.5-1 g, for children 0.25-0.5 g (daily dose of 25-50 mg / kg);

- extrapyramidal syndrome, caused by taking antipsychotics, the drug is prescribed to adults 0.5-1 g 3 times a day, children - 0.25-0.5 g 3-4 times a day. The course of treatment lasts 1 -3 months.

Tactics of prescribing the drug: increasing the dose for 7-12 days, taking in the maximum dose for 15-40 days and gradually reducing the dose until canceled within 7-8 days. The break between the course of therapy is from 1 to 3 months.

In the conditions of long-term treatment, simultaneous administration of the drug with other nootropic and stimulating agents is not recommended. Taking into account the nootropic effect of the drug, it is preferably administered in the morning and afternoon.

Side effects:

The drug is low in toxicity and well tolerated.

Allergic reactions (rhinitis, conjunctivitis, skin rashes) are possible. In this case, the drug is canceled.

Sleep disturbances or drowsiness, noise in the head - these symptoms are usually short-lived and do not require withdrawal of the drug.

Overdose:

Increased symptoms of side effects.

Treatment: Activated carbon, gastric lavage, symptomatic therapy.

Interaction:

Prolongs the effect of barbiturates, enhances the effects of anticonvulsant drugs, as well as the means that stimulate the central nervous system, enhances the action of local anesthetics (procaine).

Prevents side effects of phenobarbital, carbamazepine, antipsychotic drugs (neuroleptics).

The action of gopantenic acid is enhanced in combination with glycine, etidronic acid.

Special instructions:

In the conditions of long-term treatment, simultaneous administration of the drug with other nootropic and CNS-stimulating drugs is not recommended.

Packaging:

For 50 tablets in cans of polymeric with a barrier neck complete with a pull-on lid with a control of the first opening or in cans of polymer with a screw neck complete with a screw cap.
